Hybrid Seed purity testing: Molecular protocol

  • A lab-based molecular protocol that’s validated to match the GOT data.
  • Tests both Genetic purity and trait purity (for transgenic plants)
  • The fast and accurate protocol: up to 500 lots tested per day with only ±0.5% error rate
  • 100 - 400 seeds tested per lot and
  • Economically viable

Molecular Characterization of Lines / Hybrids / transgenic plants

Development of Trait-specific / Variety-specific Markers

  • Identify plants with desired traits based on the presence of marker(s).
  • Highly polymorphic PCR-based markers (STR or SNP based) that can distinguish parental lines, hybrids and varieties from each other.
  • The markers can be developed Trait-specific or Variety-specific based on the requirement.
  • Trait-specific markers are validated within segregating populations while Variety-specific markers are developed faster.

Screening for hybrids / Transgenic Plants

  • Screening for transformants / hybrids using transgene-specific / trait-specific markers.
  • Fast, reliable and accurate testing protocol.
  • Screening of segregating population using trait-specific markers

Transgene Copy no. detection

  • Real-Time PCR based protocol; fast, affordable and efficient estimation of copy number
  • Comparative data using house-keeping gene controls
  • Data is analyzed using software and delivered with primer sequence, raw Ct value and the copy number information for all samples.

Event Characterization Studies: Point of Integration

  • Molecular protocol to detect flanking sequences of transgene(s) integrated within the host genome
  • Genomic DNA sequence data corresponding to the flanking region of the transgene will be provided
  • Can be taken up for the studies once the copy no. is known
  • 2 weeks’ protocol and up to 10 samples can be screened together.

Development of Zygosity Test Protocols

  • Can be taken up once the point of integration is known
  • A rapid and high-throughput PCR based technique to test zygosity status of a transgenic line.
  • PCR Profile would indicate Homozygous / hemizygous or null status of the transgene allele in the transgenic plants.

Transgene Expression Profiling: Real-Time PCR Based

  • Real-Time PCR based protocol; fast and efficient 
  • Comparative data using house-keeping gene controls
  • Data is analyzed using software and delivered with primer sequence, raw Ct value and the copy number information for all samples.

Development of Plant Transformation Constructs

  • Development of plant transformation constructs with a choice of promoters, terminators, codon-optimized genes; all assembled within a plant transformation vector backbone.
  • Sequence-confirmed clones delivered.

Cloning, Expression and Purification of Bioactive Transgene Proteins

  • Recombinant proteins expressed / purified in bacterial expression vectors.
  • The recombinant proteins solubilized and re-folded following multiple re-folding protocols.
  • All refolded proteins are assayed for biological activity.
  • The refolding projects are also taken up on risk-sharing basis.

Detection and Quantitation of Recombinant Protein in Transgenic Plants

  • The recombinant / transgene proteins are detected and quantified (semi-quantitatively) within a tissue using ELISA based protocols.
  • Antibodies, secondary antibodies and other reagents required provided separately or as a kit

Genotyping / Molecular Barcoding Services:

Development of microsatellite library

  • Developed a series of highly polymorphic STR markers to genotype an organism, population or species and to understand its population genetics, conservation biology, and evolutionary biology aspects.
  • Sequence data of STR markers, specific primers and PCR conditions provided

Genotyping by SSR / STR analysis

  • Determine the genetic signature or genotype of organisms, populations or species based on naturally occurring variations using polymorphic STR markers.
  • A highly optimized protocol for fast and reliable STR genotyping by PCR / multiplex PCR assay for known microsatellite loci in any organism
  • GeneMapper-based high-resolution capillary data for all loci provided.

Microsatellite-based mapping of germplasms

  • Identification of STR markers linked to a qualitative / quantitative trait loci

Barcoding / Genotyping of germplasms by SNP-typing

  • Reproducible, fast and cost effective method to genotype / authenticate the plant varieties.
